The Barnabites commissioned San Carlo ai Catinari after their earlier church, San Biagio all'Anello, became too small. Construction began in September 1611. The church honors Saint Charles Borromeo, canonized in November 1610, and was the first church in Rome dedicated to him. The name refers to the wooden basin makers who worked in the area.

Rosato Rosati drew the main design between 1612 and 1620, and Giovanni Battista Soria designed the travertine facade, built from 1635 to 1638. The article calls the style early Baroque, with a late Baroque chapel by Antonio Gherardi. The dome is among the largest in Rome.

Domenichino frescoed the dome pendentives with the Cardinal Virtues between 1627 and 1630. The church is run by the Barnabites and serves as a titular church.
